WEEK 6

This week marked the half way point of the internship! I can't believe 5 weeks have gone by so quickly. In order to stay on track for our final presentation, all the interns gave a brief presentation about what we have accomplished so far and what we hope to accomplish in the next five weeks. I thought everyone did a great job and I enjoyed seeing the variety of projects interns are working on and how some of them fit together. I have attached my powerpoint slides from my presentation.

As far as my research goes, this week has been filled with highs and lows. Working with the new cryptophyte rubisco primers is very touch and go.... sometimes it works and sometimes it doesn't and we don't really know why. Yesterday I was so excited when I saw a DNA band in the PCR products for the 10X and 20X template dilutions. However this excitement did not last long because today the 10X template dilutions did not work. Right now a 20X template dilution is in the PCR machine so I am crossing my fingers it works!!! This process is proving very frustrating.

This week I have also been doing lots and lots of cloning. I am preparing the rest of the ETM samples from the May 2009 cruise to be sent to Washington University in St. Louis for sequencing.

We received the sequencing results from the primate center for the samples using the new Myrionecta rubra primer and the ITS primer and unfortunately these did not align with any Myrionecta sequences in the NCBI database or any sequences from Lydie's previous samples. So this lack of alignment combined with the primers with a mind of their own have led to a frustrating week. On the bright side, the new Myrionecta primer arrived today so I am anxious to try it out next week!
