More Experiment with Iron at Different Concentration - Week 9

There’s only one more week left for this internship. Everyone in the lab was so busy. It could be that everyone was wrapping up their experiments. This week I repeated the experiment from last week but with different concentration levels of Iron: 1.8 and 9 uM Fe. The measurements were the same: catalase activity, hydrogen peroxide concentration, the presence of pyoverdine, dissolved and particulate manganese (Mn), manganese oxide (MnOx), and the culture density.

On Monday, I measured the filters from last week’s experiment for MnOx through LBB assay and particulate Mn through formaldoxime assay.

On Tuesday, our lab had a practice presentation for the interns. I gave my presentation in our lab meeting and people gave me opinions on how to improve my PowerPoint. I’m so glad the interns could do the practice presentation a week ahead. This allowed me to make necessary changes to my PowerPoint and practice my presentation.

After the meeting, I prepared some minimal medium for this week’s experiment, calculated the amount of Fe needed for each sample, and grew Pseudomonas putida in LB medium.  Then from Wednesday through Friday, I measured four time points total. I still haven’t had time to plot up all my data for this week. I need to measure MnOx and particulate Mn on the filters. I ran out of time on Friday, so I’m going to do it next Monday.
