Getting Started - Week 1

This is my first time doing research internship. I’m working with Matthew Jones this summer who is out in the Black Sea for the first two weeks and a half. However I’m not alone. In this situation email come in handy. We email each other for updates and questions. Plus I got a lot of help from all the people who work in the lab. This week  I did a lot of practice with leucoberbelin blue (LBB) protocol to build a calibration curve, and grew some bacteria with help from Christine. Other people in the lab also helped me and showed me where things were placed.

On the first day I met most of the interns for this summer during orientation. After that, Christine showed me how to make some medium (LB medium and Minimal Medium A) for bacterial growths. We made the LB medium for plates, so after we autoclaved the mediums, she taught me how to pour plates. Then we streaked some plates with bacterial.

On the next few days, I went over the lab instructions from Matt for the LBB protocol and the hydrogen peroxide-amplex red (H2O2 – A-red) protocol. I was suppose to practice on the H2O2 – A-red protocol first, but it was too complicated. I had to clarify some of the procedures with Matt. Since I got one time practice of the LBB  protocol on the day before he left, it was easier for me to practice with LBB first. I emailed the results to Matt after each trials and a description of what I did for the protocol to see if I did it right. The results gave me a very nice calibration curve. Then I repeated the same protocol with many different concentrations.
